Some Immunochemical Properties of Penicillenic Acid
نویسندگان
چکیده
Under mild conditions, benzylpenicillin in aqueous solution rearranges to benzylpenicillenic acid, which reacts, under suitable conditions, to form stable disulfides and mercaptides. Through reaction with native proteins or especially with proteins that are enriched in SH groups ("thiolated"), penicillenic acid forms, ma mixed disulfide linkages, stable highly substituted conjugates. These conjugates are potent antigens, inducing the formation of substantial amounts of antibodies specific for the penicillenic acid structure. The implications of these findings for penicillin hypersensitivity in man are discussed.

Penicillin hypersensitivity in man is recognized as a clinical problem of considerable magnitude (1). Although this problem has received much attention, the antigenic determinant(s) responsible are not known. The identification of the determinant(s) is important for several reasons. First, it could permit the preparation of reliable reagents for identifying hypersensitive individuals. Second, i...

Multifunctional derivatives of penicillenic acid are effective elicitors of wheal-and-erythema skin responses in humans allergic to penicillin. Of the effective derivatives, penicilloyl-polylysines are particularly attractive as skin test reagents because they appear to be incapable of inducing antibody formation. The skin responses are specifically inhibitable in most instances by homologous u...

RECENT investigations have demonstrated the importance of the benzylpenicilloyl group as an antigenic determinant in benzylpenicillin (PG) hypersensitivity (Levine and Ovary, 1961) (Fig. 1). It has been suggested that this antigen is formed by the conjugation to protein of the highly reactive intermediate, benzylpenicillenic acid (Fig. 1, B). A number of conventional penicillins derived from 6-...
